    Enterohemorrhagic Escherichia coli colonization of human colonic epithelium in vitro and ex vivo

    Enterohemorrhagic E. coli (EHEC) are important foodborne pathogens causing gastroenteritis and more severe complications such as hemorrhagic colitis and hemolytic uremic syndrome. Pathology is most pronounced in the colon, but to date there is no direct clinical evidence showing EHEC binding to colonic epithelium in patients. In this study, we investigated EHEC adherence to the human colon by using in vitro organ culture (IVOC) of colonic biopsies and polarized T84 colon carcinoma cells. We showed for the first time that EHEC colonized human colonic biopsies by forming typical attaching/effacing (A/E) lesions which were dependent on EHEC type III secretion (T3S) and binding of the outer membrane protein intimin to the Translocated intimin receptor (Tir). A/E lesion formation was dependent on oxygen levels and suppressed under oxygen-rich culture conditions routinely used for IVOC. In contrast, EHEC adherence to polarized T84 cells occurred independently of T3S and intimin and did not involve Tir translocation into the host cell membrane. Neither colonization of biopsies nor T84 cells was significantly affected by expression of Shiga toxins. Our study suggests that EHEC colonize and form stable A/E lesions on the human colon which is likely to contribute to intestinal pathology during infection. Furthermore, care needs to be taken when using cell culture models as they might not reflect the in vivo situation

    Tests of Thermal-electric De-icing Equipment for Propellers

    Flights were made in natural icing conditions at the NACA Ice Research Project, Minneapolis, Minn. to test several designs of thermal-electric propeller de-icing blade shoes and a hub-generator design. It was found that a minimum average unit power of 2.5 watts per square inch of blade-shoe area would protect the propeller blades at the test conditions. The most satisfactory blade shoe of the three designs tested extended to the 20-percent-chord point and to 90 percent of the blade radius. A concentration of heat in the leading-edge region of this shoe was found to reduce the power input necessary for satisfactory de-icing.     Seagrass Distribution in the Pensacola Bay System, Northwest Florida

    Aerial surveys of seagrass coverage in the Pensacola Bay system (PBS) have been conducted during 1960, 1980, 1992 and 2003. This report summarizes the results for the 2003 survey and compares the results to those previously reported for other surveys. The estimated coverage of seagrass for the PBS during 2003 was 1,654 ha. Continuous and patchy coverages ranged from 0 to 684 ha and 11 to 543 ha, respectively, for five PBS subsystems. In 2003, the majority of seagrass coverage occurred in Santa Rosa Sound (76%). Declines in total coverage occurred for East Bay (93%) and Escambia Bay (75%) whereas increases were observed for Pensacola Bay (32%) and Santa Rosa Sound (8%). The approximate 9% decline (about 160 ha or 395 a) in total coverage since 1992 represents an estimated 7 to 8 million dollar loss in ecological services. The changes in coverage are likely due to naturally occurring and anthropogenic factors but it is not possible to differentiate the relative contributions of these factors alone and in combination on seagrass distribution. The ability of seagrasses to exist long-term in Florida’s fourth largest estuarine system is uncertain due to the adverse effects of rapid urbanization in the watershed. Active resource management which includes more frequent in-situ monitoring and aerial assessment and the availability of relevant water and sediment quality criteria protective of submerged aquatic vegetation are needed to prevent future declines
